New publication alert – lipidomic analysis of articular cartilage highlights changes in lipid profiles associated with osteoarthritis Osteoarthritis (OA) is not just about cartilage wear and tear—it is a complex, multifaceted disease. In our latest study, published in Annals of the Rheumatic Diseases, we uncover a previously underappreciated player in OA pathogenesis: lipid dysregulation in articular cartilage. 🔬 Key Findings: ✅ We profiled 573 lipid species in human cartilage and found striking differences between samples from OA and non-OA patients. These lipid shifts were validated in a mouse model of OA, strengthening their clinical relevance. ✅ Among the dysregulated lipids in OA chondrocytes, strikingly, we observed a reduction in odd-chain fatty acid C17:0. This pointed us towards discovering that odd-chain synthesis enzyme HACL1 is deficient in OA chondrocytes. ✅ Loss of HACL1 and accumulation of 2-hydroxy C18:0, precursor of C17:0, leads to a catabolic shift in the chondrocyte, contributing to cartilage breakdown. 🧐 Why does this matter? For years, lipids have been largely overlooked in OA research. We have long known that obesity, diabetes, and metabolic syndrome, conditions associated with a dysregulated lipid metabolism, increase OA risk. But these conditions may not just add stress to joints, and lipids appear to have fundamentally roles in cartilage biology. Our findings suggest that targeting lipid imbalances could open new therapeutic avenues in OA—a step toward disease-modifying interventions. 🚀 Looking ahead: Could interventions restore lipid homeostasis in cartilage? Could dietary or pharmacological strategies targeting lipid pathways improve the course of OA?
