🌟 See how the #NikonBioImagingLabUSA can apply #SpatialBiology tools to uncover new insights in #CancerBiology. 📄 In this poster we presented at the 2024 Society for Immunotherapy of Cancer (SITC) conference, we detail a multimodal approach for imaging and analysis of immune cell markers, immune checkpoint blockade markers, and structural markers in a non-small cell lung cancer #NSCLC patient sample. 🔬 With this method, one section is used for multiplex immunofluorescence with the #LunaphoreCOMET system, traditional H&E staining/imaging, and second harmonic generation #SHG #Imaging with the Nikon AX R MP #Multiphoton #Microscopy system. NSCLC post 2/n 🔬 We imaged a non-small cell lung cancer #NSCLC sample prepared using a 16-plex antibody panel on the #LunaphoreCOMET system at the #NikonBioImagingLabUSA. We aimed to visualize immune checkpoint blockade markers and immune cell markers in an NSCLC sample, along with some structural markers. 📖 What did we learn from the expression of immune checkpoint and other markers in NSCLC? ▶️ Apart from the widely studied immune checkpoint inhibitors (ICI) PD-1 and PD-L1 there is expression of other ICI markers. ▶️ VISTA and IDO1 are expressed in spatially distinct regions within the tissue. IDO1 is expressed on a subpopulation of PanCK+ epithelial cells, and its intensity varies across the epithelial cell population in which it's expressed. ▶️ PD-L1 is expressed on CD4+ T cells and PanCK+ epithelial cells. The PD-L1 intensity on T cells was 2.5-fold higher than on epithelial cells. ▶️ Natural Killer cells interact with VISTA+ positive cells. ▶️ Extensive remodeling of the lung tissue during NSCLC invasion, as seen by expression of a-SMA. ▶️ Presence of immature and mature tertiary lymphoid structures (TLS). 🛑 Immune checkpoint blockade markers PD-1: Helps in regulating immune responses and maintaining immune tolerance. It is expressed on T cells, B cells, monocytes, and dendritic cells. Pembrolizumab and Nivolumab are PD-1 inhibitors approved for NSCLC. PD-L1: Helps in regulating immune responses and maintaining immune tolerance. It is expressed on CD4 T cells. It is upregulated in epithelial cells during EMT, often associated with cancer progression. Durvalumab is a PD-L1 inhibitor approved for NSCLC. IDO1: Expressed in tumor cells in NSCLC, leading to an immunosuppressive tumor microenvironment, and myeloid cells. Various inhibitors are in clinical trials for use in combination with PD-1 inhibitors or other therapies. VISTA: Expressed in tumor cells in NSCLC, leading to an immunosuppressive tumor microenvironment, myeloid cells, T cells, and tumor-infiltrating lymphocytes. NSCLC Post 1/n 🔬 The video below features data from a non-small cell lung cancer #NSCLC sample prepared with a 16-plex antibody panel and imaged using the #Lunaphore COMET system at the #NikonBioImagingLabUSA. 👀 We visualized a range of immune cell markers, immune checkpoint blockade markers, and structural markers. 💪 Immune Cell Markers: CD3: Expressed on all T cells CD4: Helper T cells that interact with antigen presenting cells and help activate other immune cells CD8: Cytotoxic T cells or Killer T cells. They identify and interact with infected or abnormal cells releasing Granzyme B to induce apoptosis or programmed cell death CD20: Expressed on B cells CD56: Natural Killer (NK) cells Granzyme B: Present in NK cells and cytotoxic T cells. These cells make contact with and release Granzyme B granules in diseased cells, initiating cell death 🚧 Structural markers: CD31/PECAM-1: Expressed in endothelial cells lining blood vessels and immune cells a-SMA: Expressed by myofibroblasts and smooth muscle cells. In cancer, it's highly expressed in myofibroblasts playing a role in tissue remodeling PanCK: Detects multiple cytokeratins expressed in epithelial cells E-Cadherin: Cell adhesion molecule primarily expressed in epithelial cells 🛑 Immune checkpoint blockade markers PD-1: Helps regulate immune responses and maintain immune tolerance. It's expressed on T cells, B cells, monocytes, and dendritic cells PD-L1: Helps regulate immune responses and maintain immune tolerance. It's expressed on CD4 T cells.
