📢 Scientific Poster Our poster, "Cell Culture Process Development to Reduce High Mannose Glycans in mAb Production" was presented at Chinese Antibody Society conference. Cell culture process development serves as the crucial link in biologics CMC. MAN5 glycan is known to potentially affect the safety and efficacy of biopharmaceuticals. AsymBio’s process development team developed a new and effective cell culture approach which can significantly reduce MAN5 content by up to 78% for a molecule with a high starting MAN5 level of over 30%. The approach demonstrates effectiveness on MAN5 reduction, which can be generally applied for different IgG molecules and numerous projects. Yunnan Baiyao (YNBY) is a famous prescription for its effects on traumatic blood stasis in China. However, the side effects of YNBY including anaphylactic shock, arrhythmia and renal failure hindered its further development due to Aconite kusnezoffii Radix. (caowu) in YNBY. The purpose of our research is to study the safety of YNBY compatibility of caowu for 4 weeks on SD rats by oral administration. In this study, we determined the toxic effects on rats by conducting examinations of histopathological and clinical physiological and biochemical indicators. Next, we prospectively analyzed known metabolites using an untargeted approach in serum of SD rats administrated with YNBY and NCY (caowu replaced by excipient from YNBY) respectively for four weeks by mass spectrometry analysis. As a result, we found caowu in YNBY had no toxicity on rats according to general toxicological evaluations. It was no obvious difference between administration and control groups on AKP, GOT, GPT, BUN, CK, MDA and LDH, except Na+/K+-ATPase (ATP) and SOD index. NCY had a greater influence on rats than YNBY according to metabolism. BMKMANU S1000 Spatial Transcriptomics Case Study - Cucurbitaceae Plants Utilizing single-cell level spatial transcriptomics technology (S1000) to analyze samples from Cucurbitaceae plants, transcript data from 21,904 cells were obtained. Traditional cell segmentation methods often mistakenly identify large-diameter peripheral cells as voids and may find it challenging to distinguish cells of different sizes effectively.  However, our S-series multi-scale cell segmentation technology allows for accurate segmentation of these large-diameter peripheral cells in tissues, producing results that align closely with bright-field toluidine blue (TB) staining images. #BMKMANU #BMKGENE #cellLevel #sptatialtranscriptomics

Two-pore domain K+ (K2P) channels are a family of four-pass transmembrane K+ channels that dimerise, as homomers or heteromers, to form a functional K+ channel complex capable of regulating membrane potential through a background K+ conductance. K2P channels have widespread tissue expression, and within the nervous system they play a key role in membrane hyperpolarisation and the regulation of neuronal excitability. Moreover, these channels are implicated in a range of neurological disorders, such as pain, depression, epilepsy, mental developmental disorders, and migraine, and therefore represent attractive drug targets for these conditions. We report the development and optimisation of a TREK-1 functional assay using the Qube 384, an automated patch clamp platform capable of supporting high-throughput screening. The assay was optimized to identify both activators and inhibitors on the same plate, providing key mechanistic data for high value, limited supply screening libraries such as venom fractions used in this study.
