Our recent industry meeting inspired me to share my experience with the clinical trials budget and contract process. As always I try to provide some recommendations on how we can improve the process. Whenever I must read something with or about AI - and there is currently a lot of marketing around it, please note that the REGULATION (EU) 2024/1689 OF THE EUROPEAN PARLIAMENT AND OF THE COUNCIL of 13 June 2024 - (Artificial Intelligence Act) is in operations since 1. August 2024. This regulation applies to any system / machine / equipment supplier in the European Union and to suppliers and service providers outside of EU selling AI in EU. If we want to use AI in GXP (MedDev, GMP, CGMP in EU, GLP, GCP, GDP, GVP) this regulations applies and corresponding harmonized regulations. "How successful are AI-discovered drugs in clinical trials? A first analysis and emerging lessons" A paper from the Boston Consulting Group (BCG) published in Elsevier's Drug Discovery Today As far as i know, this is the first paper looking (holistically) at "AI-developed" drug vs drugs developed using a traditional workflow. 🌟 "In Phase I we find AI-discovered molecules have an 80–90% success rate" -> much higher than industry average 🌟 "In Phase II the success rate is $40%, albeit on a limited sample size, comparable to historic industry averages" It's still too early to judge, but it's good to have this paper to clarify what are the metrics as of 2024

China’s 15% share of world’s clinical trials in 2023 remains less than that of W Europe at 25% and N America at 23%, however, this represents an increase of 57% since 2019. During the same period the proportion of trials being conducted in W Europe declined by 21% and E Europe’s share declined by 33%. The proportion of trials being run in N American increased by 17% from 2019 to 2023.

Interesting new paper in Nature Reviews Drug Discovery on R&D costs and R&D effectiveness across the drug majors. But as far as I can see, R&D costs are calculated in way that does not reconcile with what is implied by company accounts (which would give costs that are maybe >= 2x higher). European regulatory authority adopts a positive opinion for an update of the Wegovy® label to reflect risk reduction of major adverse cardiovascular events Bagsværd, Denmark, 25 July 2024 – Novo Nordisk today announced that the European Medicines Agency’s (EMA) Committee for Medicinal Products for Human Use (CHMP) has adopted a positive opinion for an update of the Wegovy® (semaglutide 2.4 mg) label to reflect data from the SELECT cardiovascular outcomes trial, demonstrating a risk reduction of major adverse cardiovascular events (MACE) including cardiovascular death, non-fatal heart attack (myocardial infarction) or non-fatal stroke in adults with established cardiovascular disease (CVD) and either overweight or obesity (initial BMI ≥27 kg/m2) without diabetes.
