The #biosecure act is bad legislation and anti-competitive and will force US #biotech companies to abandon strong and honest #CDMO partners without making us more secure. Longstanding, productive drug-discovery collaborations will inevitably be compromised by an ill-conceived initiative — all for short-term political gain. Patients in need of innovative medicines are the ones that will lose out; I urge leaders in the #biotech industry to stand up and be counted in opposition to this proposed legislation.

I am super excited about and proud of what the Arrakis team has accomplished here. All the more so because of the sweat, talent, creativity, persistence, resilience, and orthogonal thinking it took to make this happen. While the DM1 program itself is new, it is the culmination of seven years of effort to build and optimize the physical and intellectual infrastructure needed to drug RNA with small molecules. It's been a long, occasionally frustrating, often exhausting, but always exhilarating journey. It's also, amazingly, the longest job I've held. It's been an honor. I am grateful to Arrakis employees past and present, our investors, our partners Roche and Amgen, our advisors, and everyone who has helped us one way or another. So what has the team accomplished here? In short, we have identified and optimized multiple series of drug-like small molecules that directly target the precise molecular genetic cause of myotonic dystrophy, type 1 (DM1). This devastating disease is caused by expansion of CTG repeats in the gene for DMPK, which are transcribed into mRNA carrying long extended arrays of CUG repeats. These RNAs aggregate in the nuclei of affected cells and sequester splicing factors, causing global perturbations in splicing involving dozens of genes. The impact of these aberrant splicing events on cellular proteins leads to physiological defects in multiple organs and tissues. The data that Jennifer Petter will present today establish that the Arrakis compounds specifically recognize and bind the CUG repeat - we have beautiful X-ray structures that show us precisely how they do that. They bind that sequence selectively - over other RNA sequences and the CTG repeat in DNA. They interfere with formation of nuclear aggregates in DM1 patient-derived cells. They rescue the splicing defects in these cells. Importantly, they correct splicing in a mouse model of disease and fully reverse myotonia in these animals. And they do all these things in the familiar and comforting package of drug-like small molecules, with all the properties we've come to know and love about small molecules -- including broad distribution to address tissues beyond muscle, oral bioavailability, and a relatively simple, inexpensive, and commoditized supply chain. We believe that if we can continue to advance these molecules, we'll have an important and differentiated therapeutic option for patients who have none at all today. This is why we do what we do in our business. It's wonderful when the magic works.

40 products approved in roughly a decade. There was a 3.44-fold increase in clinical trials involving CGTs between 2004 and 2023 (102 vs. 351 trials, respectively). Five hundred products are currently in the U.S. pipeline, with 10 to 20 new CGT approvals expected in 2025.

Nice in depth (and nostalgic) analysis of the new Illumina SmolSeq (🐱) by Keith Robison. Some really insightful points, most importantly IMO that SmolSeq is artfully positioned to create some competitive turbulence for Element Biosciences: "But now, $50K gets a very highly performing machine and $100K one which will fit many startup aspirations.  AVITI is still a sweet instrument, but Illumina just made that conversation more complicated." Also, why so shy about a DX version - "But even more surprising is that Illumina didn't outline a clear roadmap for clinical versions of the MiSeq i100 [🐱] - the DX model of the MiSeq received FDA clearance. The simpler workflows and smaller footprint would seem to make MiSeq i100 [🐱] a natural to try to put into pathology labs. So why the hesitancy to outline a path to get these into lucrative clinical markets?"

Happy to see the first baby from our patent granted Fc+ TCE platform, GB261, which we licensed to Genor and partner with Genor on its development will advance into the autoimmune disease field: https://lnkd.in/gBV6HvVz Since the world’s first Fc+ TCE, Catumaxomab, showed dose dependent hepatotoxicity in patients with EPCAM-positive cancers and one patient died from acute liver failure after receiving 10ug of catumaxomab, almost all major players in the TCE field either removed or silenced Fc effector function. GB261 is the world’s second Fc+ TCE entered into clinical trials and the world’s first Fc+ TCE showed great safety, efficacy balance in human at as high as 300mg dosage. The clinical POC data suggested that Ab Studio’s computer aided design platform could resolve challenges that traditional technologies can not. We hope Ab Studio’s platforms, technologies and new assets to be outlicensed will continue to support more partners!

🎉 Huge congratulations to Orum Therapeutics for their deal yesterday! Vertex bolts on Orum's Degrader-Antibody Conjugate(DAC) conditioning agents to ImmunoGen's ADC conditioner, acquired in March 2023. But what drives Vertex to explore novel conditioner candidates?   Traditional conditioning agents are poorly tolerated and have posed significant challenges for Hematopoietic Stem Cell (#HSC) Cell and Gene Therapies (CGT), such as Vertex's Casgevy. These hurdles are now being addressed by novel, less aggressive conditioners, as above. For instance, companies like Actinium Pharmaceuticals are also advancing novel conditioning agents in this space. Thus, improved conditioning agents will play a crucial role in expanding the application of HSC CGT.   Another critical challenge in HSC CGT is effective #mobilization. Emerging CGTs often require a higher yield of HSCs than conventional HSC transplantation in cancer. When it comes to in vivo approaches, having more HSCs available in peripheral blood will be critical since ex vivo collection and engineering are not feasible. Astellas Pharma's subsidiary, Xyphos Biosciences, is partnering with Poseida Therapeutics, Inc. Therapeutics to develop two "convertible" CAR-T therapy candidates for solid tumors. The goal of this collaboration is to create a safer option compared to traditional cell therapies. Xyphos' platform and Poseida's genetic editing technology will be combined to make more effective solid tumor cell therapies.
