Available for #License: Emory University researchers have found that KRASG12V contributes to #tumor formation by inhibiting the immune response pathway mediated by JAK1. They've developed a small peptide that disrupts the interaction between KRASG12V and JAK1, leading to increased killing of cancer cells carrying KRASG12V by restoring the immune response. In a recently published Perspective, Catherine Hutchings and Aaron Sato discuss phage display technology and its impact in the discovery of novel protein-based drugs. From the abstract: Areas covered In this perspective, the authors summarize the application of phage display for drug discovery and provide examples of protein-based drugs that have either been approved or are being developed in the clinic. Michael J Mitchell et al. continue to innovate and impress with their new single step #LNP formulation technique ("click-like chemistry"). They have also found a way to lojack and track these particles to better target specific organs. "We've developed what we call an amidine-incorporated degradable (AID) lipid, a uniquely structured biodegradable molecule," Mitchell says. "Think of it as an easy-to-build custom mRNA vehicle with a body kit that informs its navigation system. By adjusting its shape and degradability, we can enhance mRNA delivery into cells in a safe manner. By adjusting the amount of the AID lipid that we incorporate into the LNP, we can also guide it to different organs in the body, much like programming different destinations into a GPS." Congratulations this talented and dedicated group of scientists Aileen Li, PhD, Jessica B., Jia Lu, Quinn Walker, Rowena Ellsworth, Hajime Hiraragi, Bijan Boldajipour, Purnima Sundar, Shobha Potluri, Gary Lee, Omar Ali, and Alexander Cheung on the publication in Scientific Reports that describes our novel Stim-R T cell activation technology. CAR T cells generated using our Stim-R technology showed improved potency, expansion and cytokine production compared to standard T-cell activation reagent control. In addition, characterization of Stim-R derived CAR T-cells revealed retention of a stem-like cell population and downregulation of exhaustion-associated gene sets.  This innovative technology is incorporated in LYL119, our next generation ROR1 CAR T cell product candidate entering a Phase 1 development in a clinical trial that will initially enroll patients with ROR1‑positive platinum‑resistant ovarian cancer or endometrial cancer. https://lnkd.in/gha5VchW

Paige's lab focuses on drug discovery, employing medicinal chemistry strategies to design and synthesize small molecule modulators of dysfunctional enzymes. Structural biology and computational chemistry are integrated with kinetic assays to study enzyme mechanisms. Additionally, his lab specializes in designing, synthesizing, and characterizing peptidomimetic inhibitors of protein-protein interactions.
