A nice overview of Alzheimer's One question-- should trials be large just because diseases are large? I think Peter Kolchinsky's philosophy (? hopefully I am not stating it wrong) which seemed quite intriguing is that there are subsets of rare disease within the common disease. Maybe in an AI/ML sense defining some of the most severe populations and then rolling it back to less severe populations if it seems to work there. (Not sure if there is a path forward for that). (Note that I say this--but it doesn't necessarily seem easy). Since Alzheimer's, for instance, seems to have such diverse behavior across different genetic populations, then populations selected may influence the trial outcomes and make signals hard to decipher. Anyway Alzheimer's is so challenging because trials are likely to take a long time. And many other factors.

Once in a while I am asked how I feel about one company or another in our competitive space having signed a nice collaboration or secured a financing round. Assuming the company is doing something "real" (there are many companies not generating anything real or exciting just PR and marketing of AI), I’m happy for the company. Why am I happy to hear such news? 1. This is not a zero sum-game. The success of one company does not imply bad things for other companies in the space. In fact, the real concern about #AI in biotech is the opposite, namely, that platform companies leveraging AI for drug development would fail to get FDA clearance in the years to come and that we will all fail. In this sense, we are all playing for the same team: demonstrating that AI-based platforms can bring a real tangible value in #Tx discovery and development is crucial, and the more successes our companies would bring, the clearer this narrative will become.  2. Before being a CEO, I am a scientist, and before that I am a human being- seeing biotechnology companies advancing their capabilities for the better of patient outcomes excites me as a scientist and as a person and I will continue to cheer for companies doing great science and engineering, even if they compete with me. 3. I truly believe in competition. I think that the only way to become great is by competing with greatness. This is true in sports, true in science, and true in business. Don’t aim to win by running away from competition, aim to win by beating the competition, and Immunai aims for the gold. 4. Immunai's UVP (unique value proposition) is truly unique: we are unlocking immune system intelligence with our AI model of the immune system on the largest clinical database of the immune system. Put more simply: we are the #ChatGPT for drug development, where we are able to interrogate the immune system to answer the drug development questions. That’s something others are not doing, and something that is incredibly difficult to build. Is Dyne Done? While Dyne's corporate website announces a positive spin on DMD clinical trial results, investors were not convinced by safety nor efficacy measures leading to stock drop. CBO, COO, and CMO now announced resignation, leaving the CEO to manage the executive team catastrophe. Regardless, the company will push forward towards registration, perhaps in hopes of receiving special benevolence from Peter Marks along the way... I'm delighted to share news of this new AI-focused drug discovery company, Xaira, built on foundational technology from David Baker and led by Marc Tessier-Lavigne. The company applies AI tools to unlock opportunities in drug discovery for aspects of biology that we believe are not readily obtainable with conventional methods. I co-led NEA's investment in Xaira, along with my NEA partner and the firm’s co-CEO Mohamad Makhzoumi, and I will be joining Xaira’s board. Our enthusiasm for Xaira based on our belief that applying AI and large language models to biology holds immense promise and is still in its early stages. Currently, the application of AI to biology relies on model architectures borrowed from the tech world. A lack of well-curated clinical training sets hinders building and training these models effectively. This limits the advancement of AI in solving biological problems compared to other fields. Xaira aims to bridge this gap. We now have a clear understanding of the data needed to answer fundamental biological questions. By iterating on leading life science models and using focused perturbation data generated through our large-scale experiments, Xaira will generate data specifically suited to building advanced model architectures that are optimized for biology -- and hopefully, unlock therapeutic opportunities for patients who are not being optimally served by existing therapeutics.
