Excision of reprogramming transgenes improves the differentiation potential of iPS cells generated with a single excisable vector
Stem Cells
First author. In this study, we created an efficient method for the generation of transgene-free iPS cells and demonstrated the potential beneficial effects that result from elimination of integrated reprogramming factors.

Generation of transgene-free lung disease-specific human induced pluripotent stem cells using a single excisable lentiviral stem cell cassette
Stem Cells
First co-author. Here we developed a humanized version of our single lentiviral "stem cell cassette" vector to accomplish efficient reprogramming of normal or diseased skin fibroblasts obtained from humans.

A single Lentiviral Vector System for Induced Pluripotent (IPS) Stem Cells Derivation
Issued US 2011/0236966 A1
The present invention is based on the discovery that a single lentiviral vector expressing multiple individual transcription factor proteins from a single multi-cistronic mRNA can reprogram a fibroblast cell to a stem cell-like cell. These reprogrammed induced pluripotent stem (iPS) cells are pluripotent. Additions of the Cre-LoxP sequences into the single lentiviral vector facilitate excision of the vector after reprogramming in achieved.
