CELLxGENE really is a fantastic tool and an obligate-type resource to realizing the potential of the #singlecell & #spatialbiology. One of the things that has excited me the most about maximally unbiased multiomic data generation using techniques like #CITESeq (& now intracellular CITE-Seq) has been the idea that it could enable single cell researchers to venture beyond projection of their proteomic and transcriptomic results through curated lists to directly integrating those results with the wealth of similar data that preceded- just as we do with GEO/SRA. This dream is now materializing thanks to the work of those at Chan Zuckerberg Initiative, Human Cell Atlas 10x Genomics and researchers across the globe (Sarah Teichmann FMedSci FRS, @Aviv Regev, Nir Yosef, Aaron Streets, Rahul Satija and Nathan Salomonis to name a few). Context is key- in a vacuum, results may appear more or less interesting than they actually are - which is costly to our collective progress. While advances w/ integration has been fantastic on the RNA side, we still have some work to do with proteins at scale. Proteintech Genomics looks forward to being a part of the solution! This event was fantastic. We had a terrific group of educators and students who discussed the most pressing issues facing computational biology education: 🎓 How do we determine a core set of competencies for computational biology? 🎓 How do we address the divide between academia and industry in computational biology, given how fast things move in our field? 🎓 How do we make a more diverse and welcoming field despite the huge amount of prerequisite knowledge often needed in order to succeed?

DNA hydrogels can be a promising candidate for molecular carriers enabling precise encapsulation of specific cargo molecules and stimuli-responsive release at the target. However, they are vulnerable to nucleases making them easily degraded in a physiological environment. In this work published in ACS Applied Materials and Interfaces, we propose a simple way of controlling the enzyme resistance of DNA hydrogels by using cation-mediated condensation and expansion. We found that DNA hydrogels condensed by spermine are highly resistant to enzymatic degradation (important for cargo protection) and become degradable again if expanded back to their uncondensed state by sodium ions (useful for cargo release). We expect these controllable condensation, expansion, and degradation of DNA hydrogels would pave the way for the development of DNA hydrogels as an effective molecular carrier.

With the revived emphasis on the development of mAbs targeting HBsAg for treating chronic hepatitis B and D, this paper discusses the challenges, potentials, and provides a comprehensive overview of past and ongoing clinical trials. This aligns with two projects in our lab, collaborating with Bluejay Therapeutics (BJT-778) and Vir Biotechnology (Vir-3434, Tobevibart), assessing mAbs in clinical trials. Ratna Vadlamudi, PhD, had a long collaboration with other University of Texas System schools to discover novel drugs to treat breast cancer. During the lead optimization process of designing a drug candidate, Vadlamudi and his team discovered a small molecule that was highly active against solid cancers — ERX-41. “We made the serendipitous discovery that this small molecule had robust activity against multiple triple-negative breast cancer molecular subtypes,” Vadlamudi said. Our team is proud to present our most recent publication: 👉 For patients with metastatic colorectal carcinoma, limited RAS pathway testing is standard of care for detection of primary resistance to anti-EGFR mAb therapy. 👉 We empirically evaluate the outcomes associated with a broader set of alterations that would biologically anticipate resistance (ERBB2, RTK, and RAS/PI3K) and found that empirical outcomes associated with these broader alterations similarly associates with limited clinical benefit from anti-EGFR therapy. 👉 Evaluating post-EGFR therapy specimens, we observe a greater prevalence in these alterations as well, temporally consistent with being resistance alterations. 👉 Because these alterations are not included in standard PCR panels, the results suggest greater utility for NGS testing at baseline for clinical decisions around anti-EGFR vs. anti-VEGF use.
