Exciting updates from Bristol Myers Squibb on #mezigdomide (#CC92480) in combination with tazemetostat and dexamethasone for multiple myeloma! Mezigdomide, a potent CELMoD under investigation, has shown promising preclinical results, being significantly more potent than #iberdomide and other #IMiDs. In the CA057-003 trial (#NCT05372354), 13 patients who had undergone a median of 5 prior lines of therapy received #mezigdomide, #tazemetostat, and #dexamethasone. The overall response rate (ORR) was 53.8%, with higher mezigdomide doses yielding an ORR of 71.4%. The combination demonstrated a manageable safety profile, with grade 3/4 TEAEs occurring in 69.2% of patients. Notably, mezigdomide rapidly degrades Ikaros and Aiolos proteins, inducing apoptosis in multiple myeloma cells and stimulating the immune system. The future of CELMoDs like mezigdomide looks promising as they continue to show high synergy with traditional partners and advanced treatments. Bristol Myers Squibb has queued a Phase 1 study of Orum Therapeutics’ ORM-6151 (BMS-986497), a first-in-class, anti-CD33 antibody-enabled GSPT1 degrader for the treatment of patients with acute myeloid leukemia or high-risk myelodysplastic syndromes. The study will enroll an estimated 35 patients and is due to start in late May 2024. Orum Therapeutics has developed a new class of ADC payloads, called neoDegraders, to specifically degrade intracellular target proteins within cancer cells via the E3 ubiquitin ligase pathway. The Cancer Research Institute and its Immunotherapy Platform Study in Platinum-Resistant High-Grade Serous Ovarian Cancer (IPROC) Drug Selection Committee (DSC) have selected LOQTORZI (toripalimab-tpzi), anti-PD-1 antibody, to explore in combination with ENB-003, a first-in-class small molecule inhibitor of endothelin B receptor (ETBR), for the treatment of drug-resistant cancers in the IPROC platform study. Endothelin B receptor is implicated in tumorigenesis and tumor immune suppression for several solid tumors, including melanoma, ovarian, and pancreatic cancers. “There is a large body of clinical evidence for LOQTORZI, including positive phase 3 studies published in top-tier scientific journals, showing efficacy and safety in multiple tumor types,” says Jay Campbell, Managing Director, Clinical Accelerator and Venture Fund at CRI. “This level of scientific evidence is what we are looking for when selecting immunotherapies to bring into our network, with the goal of advancing clinically meaningful solutions to patients. Bristol Myers Squibb has posted details for a Phase 3 study (NCT06646276) of BMS-986489 (BMS-986012+nivolumab fixed dose combination) in combination with carboplatin plus etoposide vs atezolizumab in combination with carboplatin plus etoposide as first-line therapy in participants with extensive-stage small cell lung cancer. Dr. Nina E. Weisser, Director of Preclinical, Multispecific Antibody Therapeutics, recently authored an article published in Drug Target Review discussing the development of trispecific T cell engagers (TriTCEs) with integrated CD28 costimulation. TriTCEs are designed to address the limitations of conventional bispecific TCEs in treating solid tumors by providing balanced T cell activation, leading to potentially more sustainable anti-tumor responses.
