Comparing localisation and release of multivesicular bodies and secretory granules in islet cells: Priyadarshini Veerabhadraswamy at Indian Institute of Science (IISc), Nikhil Gandasi at University of Gothenburg and collaborators used the tetraspanin CD63 to study the real-time kinetics of multivesicular body exocytosis and the spatial distribution of release events within individual pancreatic β-cells. Global Genes' Director of Patient Services, Shruti Mitkus Ph.D., has contributed to the recently published N-of-1 therapies roadmap publication in Nature Reviews (with IRDiRC). An N-of-1 gene therapy is an individualized treatment developed for a particular mutation that is present in a single patient (of very few other patients). In this paper overview an overview is provided of the current advances and challenges of N-of-1 therapies as well as a development roadmap. The objective of IRDiRC's N-of-1 Task Force is to connect the different N-of-1 efforts being conducted globally and to create a roadmap towards development and implementation of these therapies. Global Genes is grateful to have provided the patient perspective on genetic diagnosis and communicating effectively with families to assess perceived benefit of such treatments. #raredisease #nof1 #careaboutrare

Negar Janani, Ph.D., Yaning Liu, Troy Butler, Erin Austin and co-authors publish "A novel application of data-consistent inversion to overcome spurious inference in genome-wide association studies" in the journal "Genetic Epidemiology". DOI: 10.1002/gepi.22563 In this article, the authors, Negar Janani, Ph.D., Kendra Young, Gregory Kinney, MPH PhD, Matt Strand, John E. Hokanson, Yaning Liu, Troy Butler, and Erin Austin, propose an assumption-free model built upon data-consistent inversion (DCI), which is a recently developed measure-theoretic framework utilized for uncertainty quantification. This proposed DCI-derived model builds a nonparametric distribution on model inputs that propagates to the distribution of observed data without the required normality assumption of residuals in the regression model. This characteristic enables the proposed DCI-derived model to cover all genetic variants without emphasizing on additivity of the classic-GWAS model. Through simulations and a replication GWAS with data from the COPDGene, the authors demonstrate the ability of this model to control the Type-I error rate at least as well as the classic-GWAS (additive linear model) approach while having similar or greater power to discover variants in different genetic modes of transmission. Collaborative Framework of the Human Cell Atlas (HCA) The Human Cell Atlas (HCA) is a global collaborative effort to map every cell type in the human body as a reference for understanding health and disease. Its collaborative framework is built on the principles of openness, inclusivity, and global scientific partnerships, ensuring that the atlas benefits from diverse expertise and represents the full biodiversity of the human population. 1. Global Network of Researchers and Institutions HCA unites scientists, clinicians, and bioinformaticians from around the world, promoting cross-disciplinary collaboration. Major contributors include leading institutions such as the Broad Institute, the Wellcome Sanger Institute, and the Chan Zuckerberg Initiative. The network spans multiple fields, from cell biology to computational science, ensuring comprehensive coverage of cellular diversity across tissues, systems, and life stages. 2. Open Science and Data Sharing At the heart of the HCA is its commitment to open science. Data generated by the project are freely shared through platforms such as the Data Coordination Platform (DCP), allowing researchers around the world to access and analyze this information. This transparency accelerates discovery and innovation while promoting reproducibility. 3. Nasal Immunotherapy for Treating Alzheimer’s Disease Researchers from the The University of Texas Medical Branch, InnoSense and University “Roma Tre,” have developed a toxic tau conformation–specific monoclonal antibody-2 (TTCM2) that selectively targets pathological tau aggregates found in Alzheimer’s disease (AD) and related disorders. By loading TTCM2 into micelles and administering it intranasally (TTCM2-ms), they achieved efficient brain penetration and clearance of tau aggregates, leading to elevated synaptic proteins and improved cognitive functions in aged tauopathy mice. The clearance process is mediated by TRIM21, which facilitates the proteasomal degradation of tau aggregates. This study demonstrates the potential of nasal tau immunotherapy using TTCM2 as an effective treatment strategy for AD by targeting and eliminating intracellular tau pathology. It has become the second leading cause of death, with approximately 75% of cancer deaths transpiring in low- or middle-income countries. It causes a heavy global economic cost estimated at more than a trillion dollars per year. The most common cancers are breast, colon, rectum, prostate, and lung cancers. Many of these cancers can be treated effectively and cured if detected at the primary stage. Nowadays, around 50% of cancers are detected at late stages, leading to serious health complications and death. Early diagnosis of cancer diseases substantially increases the efficient treatment and high chances of survival. Biosensors are one of the potential screening methodologies useful in the early screening of cancer biomarkers. This review summarizes the recent findings about novel cancer biomarkers and their advantages over traditional biomarkers, and novel biosensing and diagnostic methods for them; thus, this review may be helpful in the early recognition and monitoring of treatment response of various human cancers.
