Bristol Myers Squibb has posted details for a Phase 3 study (NCT06646276) of BMS-986489 (BMS-986012+nivolumab fixed dose combination) in combination with carboplatin plus etoposide vs atezolizumab in combination with carboplatin plus etoposide as first-line therapy in participants with extensive-stage small cell lung cancer. BMS-986012 is a novel, nonfucosylated, human IgG1 antibody that binds specifically to Fucosyl-GM1, which is a monosialoganglioside with limited expression in healthy tissues and high expression on SCLC cells. In addition, five of the 23 patients achieved a complete response, or stringent complete response, after receiving P-BCMA-ALLO1 and the higher chemo dose, the company said Friday. Separately, Poseida disclosed Friday in an SEC filing that Roche did not pick up an option for another CAR-T program targeting both BCMA and CD19. Poseida’s shares $PSTX ticked down in Monday morning trading, dipping about 3%. The updates follow Poseida’s presentation at the American Society of Hematology’s annual meeting last December. The company said then that data were “highly dependent” on patients’ cyclophosphamide regimens. Patients who took lower levels of cyclophosphamide before receiving P-BCMA-ALLO1 infusions did not respond to the CAR-T. Those results were recorded after just four weeks, raising questions about the durability of the therapy. The newest results come from a trial arm that Poseida said has an “optimized” level of cyclophosphamide at 750 mg/m2, allowing the chemo to clear out enough space in patients’ bone marrow for the CAR-T to have an effect. Median follow-up was 3.5 months, according to Poseida. Cyclophosphamide is typically given at 300 mg/m2, though it can be given in higher doses in conjunction with CAR-T. Increasing cyclophosphamide doses across trial arms appear correlated with “increased CAR-T cell expansion and persistence,” according to a note that William Blair analyst Sami Corwin wrote to investors on Friday. She added that the 91% overall response rate was higher compared to the pooled ORR of 54% from all cohorts. Corwin also suggested P-BCMA-ALLO1 could have a benefit regardless if patients previously received BCMA-targeting therapies, pointing out that all nine BCMA-naïve individuals achieved a response. There were also two patients in the 750 mg/m2 cohort who were retreated with the CAR-T at the discretion of their physicians. Poseida and Roche will move forward with the 750 mg/m2 cyclophosphamide regimen in a Phase 1b expansion study that is expected to begin “shortly,” Poseida execs said Saturday on an investor call. Poseida will receive an undisclosed milestone payment for launching the trial. ASH follow-up On top of the new cohort data, Poseida also provided updates from earlier cohorts testing other levels of cyclophosphamide. In the lowest cyclophosphamide regimen of 300 mg/m2, five of 24 patients (21%) responded, though there were no complete responses. That’s up from zero out of eight responses as reported at ASH. Both trials met their primary endpoint, with a significantly greater proportion of Sotyktu-treated patients achieving ACR20 response (at least a 20 percent improvement in signs and symptoms of disease) after 16 weeks of treatment compared with placebo. $BMY announced topline results from two pivotal Phase 3 studies for an investigational treatment in adults with #PsoriaticArthritis. Additionally, the POETYK PsA-1 and POETYK PsA-2 trials met important secondary endpoints across PsA disease activity at Week 16. The overall safety profile of Sotyktu through 16 weeks of treatment in the POETYK PsA-1 and POETYK PsA-2 trials was consistent with the established safety profile of Sotyktu observed in a Phase 2 PsA clinical trial and Phase 3 moderate-to-severe plaque psoriasis clinical trials. "Psoriatic arthritis is a heterogeneous disease that causes a range of symptoms, including joint pain and swelling, as well as psoriatic skin lesions. Despite available therapies, rheumatologists continue to express a need for a safe and effective oral treatment," said Roland Chen, MD, senior vice president and head, Immunology, Cardiovascular and Neuroscience development, Bristol Myers Squibb. Bristol Myers Squibb departed a pact with BridgeBio earlier this year, AbbVie did the same with Jacobio Pharma in 2023 and Sanofi left Revolution Medicines in 2022. Merck, meanwhile, continues to have a SHP2 deal in place with Taiho and Astex. Genentech “elected to terminate the agreement without cause,” Cambridge, MA-based Relay disclosed in a Tuesday post-market SEC filing. As part of their 2020 deal, Genentech had assumed development of the oral small molecule, dubbed GDC-1971/RLY-1971 or migoprotafib. The decision was “not based on any emerging clinical safety concerns,” a Genentech spokesperson said in an emailed statement to Endpoints News. The drug went through a monotherapy study a few years ago and is in a couple of Phase 1 trials in combination with other drugs. The pair had broad ambitions for the SHP2 inhibitor across multiple solid tumor types, including as a combination partner for Genentech’s KRAS G12C candidate known as GDC-6036 or divarasib. “Genentech is committed to ensuring that patients being treated with GDC-1971 in clinical trials continue to have access,” the Genentech spokesperson said. “All patients enrolled in a trial involving GDC-1971 or combinations thereof will continue to receive study treatment as long as they meet study criteria and they and their physician agree to continue.” Relay received $75 million upfront to kick off the collaboration in late 2020. Genentech has paid about another $45 million in biobucks and R&D reimbursements since then, according to Relay’s SEC paperwork on Tuesday. Under the original pact, Relay could have received up to $695 million in milestone payments. Roche, the parent of Genentech, has broadly fine-tuned its pipeline in the past year. Genentech itself broke off a T cell therapy tie-up with Adaptimmune earlier this year, and is also reducing about 3% of its workforce, the company told Endpoints in April. “We’ve taken out 20% of our molecules that have a lower likelihood to succeed, and we’ve added new ones through partnering,” Roche CEO Thomas Schinecker said on a first-quarter earnings call with the media in April. It’s part of normal business to prioritize and “constantly reallocate,” he added. 🚀 Opinion: Chinese CDMOs and US Biopharma PR strike a precarious balance 🌍 As the House Oversight and Accountability Committee marked up the BIOSECURE Act targeting China-based biotech companies like WuXi AppTec and WuXi Biologics, I couldn’t help but draw parallels to an epic Game of Thrones saga. 🐉🦅 In March 2024, WuXi AppTec separated from BIO after the industry trade association changed its stance to support the BIOSECURE Act.
