Roles of the Chlorine and Methoxy Groups in Drug Discovery | https://lnkd.in/gpvXdSC9 Thu, Sep 26, 2024 8 AM PDT / 11 AM EDT / 5 PM CEST Small functional groups can greatly influence the interactions of a small-molecule ligand with the binding pocket of a protein or nucleic acid. Possible steps that a medicinal chemist could take to optimize a phenyl-containing hit molecule are summarized in the Topliss tree. The most important substituents that one encounters in this flowchart are chloro, methyl, and methoxy. Although the role of the methyl group has been highlighted in the literature and referred to as "magic methyl" to describe unexpectedly significant potency improvements, the roles of the chloro and methoxy groups in drugs has not been thoroughly explored. In this Flash Talk, Yoshihiro Ishihara will discuss the beneficial effects of chloro and methoxy substituents on intermolecular interactions, as well as the unique advantages and disadvantages of employing these substituents in drug discovery. RapidFire-MS was invented to address the limitations of traditional LCMS methods used in drug discovery and development. In the early 2000s, LCMS was the primary tool for analyzing large numbers of compounds in drug screening assays. However, it was relatively slow and oftentimes offered more separation than was necessary for MS analysis. To overcome long unnecessary chromatographic times, RapidFire-MS was developed as a high-throughput screening technology that could analyze large numbers of compounds in a short amount of time. The average cycle time for RapidFire was 8 seconds compared to many minutes for LC. This significant improvement in throughput was achieved by faster sampling, shorter flow paths, and bind/elute cleanup (instead of chromatographic separation). RapidFire-MS was designed to meet the demands of the pharmaceutical industry for faster and more efficient drug screening technologies. It has since become a widely used tool in drug discovery, metabolomics, and proteomics. 🚨New technical note alert🚨 This technical note describes the workflow for per- and polyfluoroalkyl substances (PFAS) characterization in cosmetics using non-targeted acquisition (NTA) with compound identification through suspect screening and diagnostic fragment ion screening. Using the ZenoTOF 7600 system, data-dependent acquisition (DDA) was used to obtain TOFMS precursor and TOFMSMS fragmentation spectra.
